What does penicillin target?

Cell wall peptidoglycan biosynthesis.

How does penicillin work?

Beta-lactam ring.  

    (Also a thiazolidine right)

Substrate analog; forms acyl-enz. intermediate.

Penicilloyle-enzyme is inactive, blocking crosslinking

Beta-lactamase cleaves beta-lactam.
